Background: Interleukin-36 (IL-36) is not a single cytokine but a group of proinflammatory cytokines that belong to the IL-1 family and are expressed in epithelial barriers such as the skin, gut, and lungs. The IL-36 group consists of three ligand isoforms: IL-36α, IL-36β and IL-36γ, which are activated when the pro-protein forms are cleaved by neutrophil derived proteases. Signaling is initiated when IL-36 binds to the Interleukin 36 receptor (IL-36R), also known as IL-1RL2, and its co receptor IL-1 receptor accessory protein (IL-1RAP). This triggers a pro-inflammatory response predominantly through the NF-κB (nuclear factor kappa-light-chain-enhancer of activated B cells) and MAPK (mitogen-activated protein kinase) signaling pathways. Dysregulated IL-36 signaling has been implicated in a variety of inflammatory diseases, most notably psoriasis where IL-36 is abundantly expressed in keratinocytes (KCs) on the skin. The field of IL-36-targeted therapies is rapidly evolving, spearheaded by the development of IL-36R antagonists. Spesolimab, FDA-approved for the treatment of generalized pustular psoriasis (GPP), is a humanized monoclonal antibody that blocks IL-36R, effectively inhibiting the activation of downstream inflammatory pathways. Additional IL-36R blocking therapies have shown promising results, with Imsidolimab currently undergoing Phase III trials for the treatment of GPP.

Biological Activity: This protein was tested for its ability to induce IL-8 secretion in A431 mouse embryonicfibroblast cells.

Description: Recombinant human IL-36 alpha, also known as IL-1F6, encompassing amino acids 6-158, corresponding to the full lenght mature protein. This construct does not have a tag. The protein was affinity purified.

Format: Lyophilized powder.

Formulation: Lyophilized from a 0.22 μm filtered solution of 50 mM Tris-HCl, 150 mM NaCl, 1 mM EDTA, 0.02% Tween 20, pH 8, OR from a 0.22 μm filtered solution of 20 mM PB, 6% Trehalose, 5mM EDTA, 0.05% Tween 80, pH7.4.Note: For SPR assay, please replace the buffer. Primary amine components (e.g., Tris, imidazole) can affect protein-coupled chips.

Purity: ≥95% by reducing SDS-PAGE.

Reconstitution: It is not recommended to reconstitute to a concentration less than 100 μg/ml in distilled water. For long term storage it is recommended to add a carrier protein (0.1% BSA, 5% HSA, 10% FBS or 5% Trehalose).

Storage Stability: Store at -20°C for 2 years from date of receipt. After reconstitution, it is stable at 4°C for 1 week or -20°C for longer (with carrier protein). It is recommended to freeze aliquots at -20°C or -80°C for extended storage.
