Specificity Statement: The antibody binds specifically to SpuE. In particular, it interacts with the hinge region that connects the two domains of SpuE.

Application Notes (Clone): The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody conjugated to a membrane penetrating peptide (MPP) could inhibit the secretion of ExoS from P. aeruginosa. The antibody conjugated to MPP was able to inhibit the cytotoxic effects of SpuE in the human epithelial cell line A549. The antibody conjugated to MPP showed a protective effect on C. elegans from P. aeruginosa infection. The crystal structure of the antibody in complex with SpuE-spermidine was determined. The dissociation constant (KD) of SpuE to spermidine and of SpuE to spermidine in the presence of the antibody was measured by isothermal titration calorimetry (Kd = 2.7 nM and 29.8 nM, respectively) (Zhang et al., 2019; PMID: 31682834).
