CloneID: 10E4
Heavy Chain modification: Fc Silent™
Antigen Long Description: The original antibody was isolated from a human naïve scFv phage display library by panning it against purified human CD9-ECL2 protein.
Buffer Composition: PBS only.
Chimeric Use Statement: This is a reformatted human IgG1 Fc Silent™ antibody, based on the original human scFv format, created for improved compatibility with existing reagents, assays and techniques.
Uniprot Accession No.: P21926
Specificity Statement: This antibody specifically binds amino acids 'KEVFDN' in the extracellular loop 2 domain (CD9-ECL2) of human CD9. This protein is an intergral membrane protein associated with integrins, which regulates different processes, such as sperm-egg fusion, platelet activation and aggregation, and cell adhesion. Extracellular vesicles (EVs) such as exosomes are important mediators in intercellular communications and play a role in cancer progression and metastasis. Exosomal membranes are enriched in endosome-specific tetraspanins like CD9. EVs have strong potential as blood-based or urine-based biomarkers for the diagnosis, prognostication, and surveillance of cancer.
Application Notes (Clone): The binding of this antibody to CD9 expressed on different cancer cell lines like 2774 cells, U031 cells, ANBL-6 cells, 293E cells was tested using flow cytometry (US8197809). The epitope characterization of this antibody for fragment of CD9-ECL2 was measured using ELISA. This antibody significantly inhibited invasion of 2774 (ovarian adenocarcinoma cell line) cells and further inhibited migration and proliferation of 2774 cells (US8197809).
