Specificity Statement: This antibody binds human CXCL10. This pro-inflammatory cytokine is involved in a wide variety of processes such as chemotaxis, differentiation, and activation of peripheral immune cells, regulation of cell growth, apoptosis and modulation of angiostatic effects. This protein plays an important role during viral infections by stimulating the activation and migration of immune cells to the infected sites. Binding of CXCL10 to the CXCR3 receptor activates G protein-mediated signaling and results in downstream activation of phospholipase C-dependent pathway, an increase in intracellular calcium production and actin reorganization.
Application Notes (Clone): The binding characterization of this antibody was done using ELISA. The binding affinity of this antibody towards human CXCL10 was determined using BIACORE and was found to be Kd= 9.2 nM. The neutralizing potential of the IgG version of this antibody on hCXCL10-induced calcium flux or cell chemotaxis was evaluated using the cell-based assays. This antibody was reported to inhibit the activity of both recombinant and native hCXCL10 in a dose-dependent manner. CXCL10 is able to oligomerize and bind to glycosaminoglycans (GAG) expressed at surface of cells such as endothelial cells. This antibody was capable of binding human CXCL10 immobilized on glycosaminoglycan (US7786268).
