Antigen Long Description: The original antibody was isolated from a naïve human single chain Fv (scFv) phage display libraries using a series of selection cycles on recombinant human IL-1R1. The scFv was later reformatted to the human IgG1 version.

Specificity Statement: This antibody binds human IL-1R1 and cynomolgus monkey IL-1R1 but does not cross react with mouse IL-1R1. This antibody binds an epitope containing amino acids in a discontinuous epitope of 33 amino acids in three regions of human IL-1R1; N123-V134, L140-K157 and K178-R180. The human IL-1R1 protein acts as a receptor for IL1A, IL1B and IL1RN.
Application Notes (Clone): The binding affinity of this antibody to human IL-1R1 was determined using kinetic exclusion assay. It was reported that the human IgG1 version of this antibody binds human IL-1R1 with a binding affinity of 3.05 pM. The human IgG1 version of this antibody shows the inhibition of IL-1β induced IL-8 release from HeLa cells. The IgG2 version was capable of inhibition of IL-1β and IL-α induced IL-8 release from HeLa cells. The binding of this antibody was capable of inhibiting binding of IL-1Ra binding to IL-1R1. This antibody was reported to inhibit huIL-1β-induced IL-8 release from CYNOM-K1 cells. The human IgG1 and IgG2 versions of this antibody was capable of inhibiting IL-6 production in human whole blood. The IgG2 version also blocked IL-1 activity (US8298533).
