Assay Conditions: A white 96-well plate was precoated with IL-1RL2 protein (MedChem Express #HY-P70603) (100 ng/well). Blocking Buffer 3 (#79743) and PBST were used for blocking and washing steps, respectively. For protein titration a serial dilution of Anti-IL-36R Antibody (#102719) at the highest concentration of 2 ng/µl (100 ng/reaction) was prepared in 1x PP-02 Assay Buffer (#82620). Diluted Anti-IL-36R was then added to the wells and incubated at Room Temperature (RT) for 1 hour. The plate was washed again and incubated with anti-Fc-HRP conjugate diluted 1:1000 in Blocking Buffer 3 for 1 hour at RT. After the final wash, ELISA ECL mix (#79670) was added to each well and chemiluminescence was read.
Background: Interleukin-36 (IL-36) is a member of the IL-1 cytokine family and includes IL-36α, IL-36β, and IL-36γ. The IL-36 receptor (IL-36R), complexed with the IL-1 receptor accessory protein (IL-1RAcP), binds to IL-36 to activate the NF-kB (nuclear factor kappa B) and MAPK (mitogen-activated protein kinase) signaling pathways. This activity plays a role in innate and adaptive immune responses, namely in psoriasis and inflammatory bowel disease. There is therapeutic potential in the development of monoclonal antibodies and inhibitors for IL-36R. For example, Spesolimab is currently undergoing clinical trials to treat generalized pustular psoriasis (GPP).
