Assay Conditions: A white 96-well plate was precoated with ITGA4-ITGB7 protein (AcroBiosystems #IT7-H52W4) (100 ng/well). Blocking Buffer 3 (#79743) and PBST were used for blocking and washing steps, respectively. For protein titration a serial dilution of Anti-ITGA4-ITGB7 Neutralizing Antibody (#102488) at the highest concentration of 10 ng/µl (250 ng/reaction) was prepared in 1x PP-02 Assay Buffer (#82620). Diluted Anti-ITGA4-ITGB7 Neutralizing Antibody was then added to the wells and the plate was incubated at Room Temperature (RT) for 1 hour. The plate was then washed again and incubated with an anti-Fc-HRP conjugate diluted 1:1000 in Blocking Buffer 2 for 1 hour at RT. After the final wash, ELISA ECL mix (#79670) was added to each well and chemiluminescence was read.
Background: Integrin alpha 4 beta 7 (α4β7) is also known as Lymphocyte Peyer’s patch adhesion molecule-1 (LPAM-1), a heterodimer composed of the integrin beta 7 subunit (ITGB7) and the ITGA44 subunit (also known as CD49d or integrin alpha 4), of interest in studies of the gastrointestinal tissue. Key interactions include surface adhesion molecules such as MADCAM1 (mucosal vascular addressin cell adhesion molecule 1) and VCAM (vascular cell adhesion molecule), as well as fibronectin. This protein has been linked to diseases such as UC (ulcerative colitis) and Crohn’s disease, amongst others. There are several approved LPAM-1 targeting therapeutics, such as vedolizumab (commercially known as Entyvio, Takeda), that aim to mitigate inflammatory bowel disease and cancer, with more clinical trials underway.
