CloneID: OX-133
Antigen Long Description: Purified MBP-gp120 (HIV-1) reduced using tris(2-carboxyethyl)phosphine (TCEP, 2.5 mM) for 20 minutes and alkylated using NEM (5 mM solubilized in PBS) for 30 minutes.
Buffer Composition: PBS only.
Chimeric Use Statement: This chimeric rabbit antibody was made using the variable domain sequences of the original Mouse IgG1 format, for improved compatibility with existing reagents, assays and techniques.
Specificity Statement: This antibody recognises cysteine-boud N-ethylmaleimide in a sequence-independent manner. It has no affinity for unmodified peptides or peptides modified with other alkylating agents such as iodoacetamide.
Application Notes (Clone): OX133 detects NEM-modified proteins and can be used as an affinity matrix to purify NEM-modified proteins from cell lysates. OX133 does not cross react with any other alkylating agent, making it a highly selective reagent for the purification of NEM-labeled proteins and potentially peptides for mass spectrometry-based analysis.
