Assay Conditions: This protein was tested using an Alpha-LISA™ binding assay. A 10 µl reaction mix containing E-cadherin in 1x PP-02 Buffer was incubated with Cadherin3, Avi-His-Tag, Biotin-Labeled Recombinant for one hour at Room Temperature (RT). Anti-IgG1 acceptor beads were added, and the reaction was incubated for 30 minutes, followed by the addition of Streptavidin-conjugated Donor beads. A-counts were measured. The net A-count signal is proportional to E-Cadherin-Cadherin3[B] binding.
Background: Cadherins are calcium-dependent cell adhesion proteins. They preferentially interact with themselves in a homophilic manner to connect cells. Cadherin3, also known as CDH3 or P-cadherin, is a type I cadherin, found in the placenta but mostly in epithelial tissues. It plays a role in cell growth and migration. It is implicated in cancer, with high levels in breast cancer being linked to poor prognosis. Soluble cadherin3 can be used as a cancer biomarker. Its role in cancer seems to be context dependent, as in some instances its level seems to correlate with suppressed tumorigenesis. Further studies will allow us to have an in-depth knowledge on the role of this protein in pathologies and to develop therapeutic approaches linked to it.
