Application: Useful for studying expression of human EGFR on the cell surface by flow cytometry.
Assay Conditions: One million Human EGFRvIII CHO K1 cells (#78145-H) were blocked and stained with 1 µg of Anti-EGFR Antibody, PE-Labeled (#102669) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. Human EGFRvIII CHO K1 cells (green) were compared to parental CHO-K1 cells (blue). Y-axis is the cell count. X-axis is PE intensity.
Background: The epidermal growth factor receptor (EGFR, ErbB-1, HER1) belongs to a family of receptor tyrosine kinases that includes three other closely related members (HER2, HER3 and HER4). It regulates cell growth, survival and differentiation. It is activated by binding to ligands like epidermal growth factor (EGF), leading to receptor dimerization and activation of signaling pathways such as RAS/MAPK (mitogen-activated protein kinase), PI3K (phosphoinositide 3-kinase)/AKT (protein kinase B), and JAK (janus kinase)/STAT (signal transducer and activator of transcription). EGFR is crucial for normal cellular functions but is often overexpressed or mutated in cancers, including non-small cell lung cancer (NSCLC), colorectal cancer, and glioblastoma, making it a key target for therapies like tyrosine kinase inhibitors (TKIs) and monoclonal antibodies.
Cross Reactivity: This antibody recognizes human EGFR. It has not been tested in other species.
Description: This anti-EGFR (epidermal growth factor receptor) antibody is a purified monospecific antibody, which is labeled with R-Phycoerythrin (PE). This antibody has been tested by flow cytometry for binding to human EGFR on the surface of Human EGFRvIII CHO K1 Cell Line (High Expression) (BPS Bioscience #78145-H). FluoSite™ is a tag-based method for site-specific conjugation of antibodies with fluorophores, ensuring the labeling site is far from the antigen-binding domain to preserve antibody functionality. This precise, consistent labeling ensures the homogeneity of the labeled antibody preparation, making it ideal for flow cytometry and other high-sensitivity applications.
