Applications: Useful for studying expression of CD14 on the cell surface by flow cytometry. For flow cytometric staining, the suggested use of this reagent is 5 µl (1 µg antibody) per million cells in 100 µl staining volume.
Assay Conditions: One million CD14 CHO cells (BPS Bioscience #78335) were blocked and stained with 1 µg of anti-CD14 antibody, FITC-labeled (BPS Bioscience #101884) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. CD14 CHO cells (green) were compared to parental CHO-K1 cells (blue). Y-axis represents the cell count, while X-axis shows PE intensity.
Background: CD14 (Cluster of Differentiation-14) is a surface antigen expressed predominantly by monocytes and macrophages. It is considered a patterned recognition receptor (PRR), which recognizes pathogen markers. Specifically, CD14 acts as a co-receptor for lipopolysaccharides (LPS) found in the outer membrane of gram-negative bacteria. Through this interaction, CD14 supports initiation of the innate immune response during bacterial infection. CD14 also contributes toward immune responses to viral pathogens such as human respiratory syncytial virus (RSV) and may amplify the inflammatory response observed in severe cases of SARS-CoV-2 infection (COVID-19). Monoclonal antibodies against CD14 are currently undergoing clinical trial as a candidate treatment to limit severe inflammatory responses common in patients hospitalized with COVID-19.
Cross Reactivity: This antibody recognizes human CD14. It has not been tested on other species.
Description: This anti-CD14 antibody is a purified recombinant antibody, which is labeled with fluorescein isothiocyanate (FITC). This antibody has been tested by flow cytometry for binding to human CD14 on the surface of CD14 CHO Recombinant Cell Line (BPS Bioscience #78335).
Format: Aqueous buffer solution.
Formulation: 8 mM Phosphate, pH 7.4, 110 mM NaCl, 2.2 mM KCl, 0.09% Sodium Azide, 0.2% BSA, and up to 20% glycerol. May contain a protein stabilizer.
Host Cell Line: HEK293
Purification: Protein A affinity chromatography from HEK293 supernatant.
Purity: ≥90%
Storage Stability: At least 6 months at 4°C. Protect from prolonged exposure to light. DO NOT FREEZE. The antibody solution should be stored undiluted between 2°C and 8°C.
