Applications: Useful for studying expression of CD3 on the cell surface by flow cytometry.
Assay Conditions: One million PBMCs (BPS Bioscience #79059) were blocked and stained with 1 µg of anti-CD3 antibody, PE-Labeled (BPS Bioscience #102009) or PE Human IgG1 Isotype Control (BioLegend #403503) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. Non-lymphoid cells were excluded from the analysis based on side scatter and forward scatter signals. Y-axis represents the cell count while the X-axis indicates PE intensity.
Background: CD3 (cluster of differentiation 3) is a complex made of CD3γ chain, a CD3δ chain and two CD3ε chains. CD3 associates with the TCR (T-cell receptor) and a CD3ζ chain to activate cytotoxic T Cells (CD8+) and T helper cells (CD4+) and form the TCR complex. It is found in T cells, during all developmental stages, and it is a typical T cell marker. Its essential role in T cell activation has made CD3 an attractive and useful molecule for the development of anti-cancer drugs. For example, CAR-T cells (chimeric receptor antigen T cells) made use of the CD3ζ domain for intracellular signaling activation.
Cross Reactivity: This antibody recognizes human CD3. It has not been tested on other species.
Description: This anti-CD3 antibody is a purified recombinant antibody, which is labeled with R-Phycoerythrin (PE). This antibody has been tested by flow cytometry for specific binding to human CD3 on the surface of PBMCs (BPS Bioscience #79059).
Formulation: 8 mM Phosphate, pH 7.4, 110 mM NaCl, 2.2 mM KCl, 0.09% Sodium Azide, 0.2% BSA, and up to 20% glycerol. May contain a protein stabilizer.
Purification: Protein A affinity chromatography from HEK293 supernatants.
Purity: ≥90%
Storage Stability: At least 6 months at 4°C. Protect from prolonged exposure to light. DO NOT FREEZE. The antibody solution should be stored undiluted between 2°C and 8°C.
Warnings: Do not freeze. Protect from light.
