Applications: Useful for studying expression of CLDN18.2 on the cell surface by flow cytometry. For flow cytometric staining, the suggested use of this reagent is 5 µl (1 µg antibody) per million cells in 100 µl staining volume.
Assay Conditions: One million CLDN18.2 CHO cells (BPS Bioscience #78533) were blocked and stained with 1 µg of anti-CLDN18.2 antibody, FITC-labeled (BPS Bioscience #101866) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. CLDN18.2 CHO cells (green) were compared to parental CHO-K1 cells (blue). Y-axis represents the cell count, while X-axis shows FITC intensity.
Background: Claudins are integral membrane proteins and components of tight junctions. The claudin-18 gene has two splice variants: claudin-18.1, which is found primarily in the lung, and claudin-18.2, which is expressed exclusively in the stomach. Claudin-18.2 is highly expressed in gastric and pancreatic adenocarcinoma and may be involved in tumor development and progression. It is an ideal target for therapy and effort has been put into the development of monoclonal antibodies against claudin 18.2, such as Zolbetuximab. Studies of claudin-18.2 function and expression patterns will continue to broaden our knowledge and application in cancer therapy.
Cross Reactivity: This antibody recognizes human CLDN 18.2. It has not been tested on other species.
Description: This anti-Claudin-18 isoform 2 (CLDN18.2) antibody is a purified recombinant antibody, which is labeled with fluorescein isothiocyanate (FITC). This antibody has been tested by flow cytometry for specific binding to human CLDN18.2 on the surface of CLDN18.2 CHO Recombinant Cell Line (BPS Bioscience #78533).
Format: Aqueous buffer solution.
Formulation: 8 mM Phosphate, pH 7.4, 110 mM NaCl, 2.2 mM KCl, 0.09% Sodium Azide, 0.2% BSA, and up to 20% glycerol. May contain a protein stabilizer.
Host Cell Line: HEK293
Purification: Protein A affinity chromatography from HEK293 supernatant.
Purity: ≥90%
Storage Stability: At least 6 months at 4°C. Protect from prolonged exposure to light. DO NOT FREEZE. The antibody solution should be stored undiluted between 2°C and 8°C.
