Notes: Recombinant prokaryotic lectin-αMan (RPL-αMan) is a P. aeruginosa lectin expressed in and purified from E. coli. It binds to glycoproteins containing Lewis a (Lea), Lewis X (LeX), or terminal α-mannose groups. RPL-αMan activity requires metal ions, therefore, all buffers must be supplemented with 1 mM calcium chloride, magnesium chloride, and manganese chloride. It is recommended to use metal ion-supplemented Tris-buffered saline (TBS), pH 7.6, to dilute RPL-αMan to the desired working concentration (≤20 µg/ml). Following dilution, incubate RPL-αMan for a minimum of 30 minutes prior to use to allow metal complexation.
