Background: HERC1 (HECT domain and RCC1-like domain-containing protein 1) belongs to the HERC family of ubiquitin ligases. It is involved in membrane traffic via its ability to bind clathrin and its guanine nucleotide exchange factor (GEF) activity for Arf and Rab. HERC1 has been found to interact with TSC2 (tuberous sclerosis complex 2) and may be involved with its destabilization.
