Background: RNF31, or RING finger protein 31, is an E3 ubiquitin ligase also known as HOIP (HOIL-1 Interacting Protein). This protein is a component of the linear ubiquitin chain assembly complex (LUBAC) and is understood to promote tumor growth via activation of the nuclear factor kappa B (NF-kB) pathway. Overexpression of RNF31 is linked to breast cancer, hepatocellular carcinoma, and melanoma. The development of RNF31 inhibitors, such as HOIPIN-8, and other drugs will improve cancer outlooks and expand our understanding of tumor progression and the NF-kB pathway.
