Background: USP15 is a deubiquitinase that associates with the COP9 (constitutive photomorphogenesis 9) signalosome (CSN) complex. It regulates the NF-kappaB pathway by deubiquitinating IkappaBalpha2. USP15 acts as a key regulator of TGF-beta receptor signaling pathway and is thought to promote deubiquitination of monoubiquitinated R-SMADs (SMAD1, SMAD2 and/or SMAD3), thereby alleviating inhibition of R-SMADs and promoting activation of TGF-beta target genes. UPS15 is linked to immune and inflammatory roles, where it can regulate the transcription of pro-inflammatory cytokines and type I interferons. It also plays a role in the TEM (tumor microenvironment). More research will shed light on the role of USP15 as a cancer therapy target.
