Application: Use as a control for studying differences between HLA-B alleles. Study T cell activation and responses to peptides presented by HLA-B*35:01.
Background: Human Leukocyte Antigen-B (HLA-B) is an MHC-I (major histocompatibility complex) heavy chain receptor, composed of HLA-B and β2-microglobulin (B2M). There are over 200 genes encoding HLA variants and this variability plays a critical role in adaptive immunity. HLA-B has been linked to autoimmune disorders and drug reactions. B2M HLA-B*35:01 has been linked to drug-induced liver injury (DILI) in response to the Kampo traditional Japanese medicine and Polygonum multiflorum (PM) Chinese herbal medicine. The understanding of the association between HLA alleles and diseases will allow a refinement of therapeutic strategies.
Description: The HLA-B*35:01 A549 Cell Line is an A549 human lung cancer cell line generated by selecting a single clone of B2M Knockout A549 cells (#82871) transduced with a lentivirus expressing B2M (beta-2 microglobulin) HLA (human leukocyte antigen)-B*35:01 driven by an EF1a promoter (#82431). This cell line has been validated by flow cytometry.
Host Cell Line: A549 is a human lung alveolar basal carcinoma cell line. Adherent epithelial cells.
Mycoplasma Testing: The cell line has been screened to confirm the absence of Mycoplasma species.
Storage Stability: Cells are shipped in dry ice and should immediately be thawed or stored in liquid nitrogen upon receipt. Do not use a -80°C freezer for long term storage.
Supplied As: Each vial contains ˃1 x 106 cells in 1 ml of Cell Freezing Medium (#79796).
