Specificity Statement: This antibody specifically binds mouse CCR2 and does not cross react with mouse CCR5 or human CCR2 and CCR5. CCR2 is a key functional receptor for CCL2 but can also bind CCL7 and CCL12 chemokines. Its binding with CCL2 on monocytes and macrophages mediates chemotaxis and migration. It regulates the expression of T-cell inflammatory cytokines and T-cell differentiation, promoting the differentiation of T-cells into T-helper 17 cells (Th17) during inflammation.

Application Notes (Clone): This antibody can be used to detect mouse CCR2 expressed on the surface of CHO cells and mouse peripheral blood leukocytes using flow cytometry. This antibody is capable of reducing the activity of murine monocyte chemotactic protein 1 in a manner similar to the well characterized anti-CCR2 antibody MC-21. This antibody can prevent the influx of monocytes in thioglycolate-induced peritonitis. This antibody is also capable of blocking the binding of mouse CCR2 with its ligand in vitro and in vivo. The blockade of CCR2 with this antibody also impairs monocytes/macrophage recruitment in vivo. This antibody can be used to determine the role of chemokine receptor CCR2 during initiation and progression of collagen induced arthritis in mice using blocking antibodies. This antibody is capable of abolishing long-term behavioral recovery after ischemic stroke and drastically decreased tissue expression of anti-inflammatory genes, including TGFβ, CD163, and Ym1.
