FLAER is a proaerolysin fluorescently labeled with Alexa® 488. FLAER represents an important part of antibody detection panels in clinical flow cytometry for detection of paroxysmal nocturnal hemoglobinuria (PNH). PNH is a disorder affecting stem cells, which is caused by improper synthesis of glycosylphosphatidylinositol (GPI) due to gene mutation. Functional GPI serves as an anchor for surface proteins on circulating cells, therefore affected cells are prone to complement-mediated hemolysis leading to several life threatening states like thrombosis, chronic kidney disease, pulmonary hypertension, end organ damage, ischemic bowel disease, hepatic failure and anemia. FLAER binds selectively and with high affinity to GPI-anchored proteins and due to negative selection enables very sensitive detection of PNH cell populations (FLAER negative cells).
