Antigen Long Description: The original antibody was generated from a phage Fab library created from the B-cells of CHIKV infected patients. The library was panned against retroviral (Gag core) and CHIKV (alphavirus capsid core) virus-like particles (VLPs) displaying the native form of CHIKV E2/E1.

Specificity Statement: The antibody is specific for chikungunya virus. The epitope was mapped to an area of E2 encompassing domains A and B, and arches 1 and 2. Chikungunya is a mosquito-borne viral disease caused by the chikungunya virus, an RNA virus in the alphavirus genus of the family Togaviridae.
Application Notes (Clone): The binding affinity of the original format of the antibody to CHIKV VLPs was measured by surface plasmon resonance (Kd= 9.3 nM). The original antibody demonstrated high reactivity with E2/E1 presented on cells, as shown by flow cytometry. The original format of the antibody was tested in a cellular neutralization assay against CHIKVL, where showed an average IC50 of 170 ng/ml. The original antibody was tested for antiviral activity using native replication-competent CHIKV viruses. In a plaque reduction neutralization test assay, the antibody could neutralize CHIKV strain 37997 with a PRNT50 value of 81 ng/ml (US9902765B2).
