Antigen Long Description: The original antibody was generated from the peripheral blood plasmablast cells from three humans immunized with vaccines containing FHbp. The natively paired VH and VL genes were expressed in Escherichia coli to obtain recombinant antibody fragments (Fabs).

Specificity Statement: This antibody binds a surface exposed epitope on N. meningitidis FHbp, a virulence factor critical for surviving of the bacteria in the human host. This antibody binds equally well to fHbp variants, including representative variants in variant groups 1, 2 or 3. FHbp protein is a bacterial surface lipoprotein that binds host (human) complement factor H (fH, gene CFH). Neisseria meningitidis is the leading global cause of meningitis and sepsis.

Application Notes (Clone): The original Fab version of this antibody binds N. meningitidis fHbp protein with a very high binding affinity of Kd= 19pM. This antibody is broadly cross reactive and binds FHbp sequence variants from groups 1, 2, and 3. In an ELISA, this antibody shows concentration dependent binding to recombinant FHbp (ID 1), which is the variant group 1 antigen (PMID: 26106082). Flow cytometry was used to identify binding of this antibody to surface of live bacteria of N. meningitidis strains serogroup B strain H44/76, which expresses FHbp ID 1 (PMID: 26106082). In functional assays, IgG version of 1A12 has bactericidal activity against strains expressing each of the three fHbp variants (PMID: 29410413; 29487324).
