Specificity Statement: This antibody binds and epitope between amino acid R1803-K1818 of the A3-C1 domains of the human factor VIII, which also contain binding site for factor IXa. Factor VIII is a glycoprotein involved in blood coagulation. Factor VIII acts as a cofactor for factor IXa which converts factor X to the activated form Xa in the presence of calcium and phospholipids. Defects in the F8 gene, which encodes Factor VIII, result in the coagulation disorder hemophilia A.
Application Notes (Clone): The scFv version of this antibody was capable of inhibiting factor VIII activity with inhibition titre of 63 BU/mg. The scFv antibody binds human factor VIII light chain with a binding affinity of Kd= 1.6 nM (PMID: 11159524). It was reported that scFv KM33 inhibits the procoagulant activity of factor VIII. In view of the similar epitope specificity of scFv KM41 and CLB-CAg A (antibody which binds a synthetic peptide comprising residues R1803-K1818), it is likely that scFv KM41 competes with factor IXa for binding to the factor VIII light chain (PMID: 11159524).
