Specificity Statement: This antibody is specific for a 79 amino acid long region between amino acids 1462 to 1540 of the full human sequence (corresponding to amino acids 1399 to 1478 in the rat homologue) in Giantin, a Golgi matrix protein.
Application Notes (Clone): The original format of this antibody (scFv) was successfully used in a Western blot and for in vitro immunofluorescence straining of Giantin in HeLa cells. siRNA knock-down of Giantin reduced TA10 staining, confirming its specificity. Furthermore, an in vivo expressed TA10-GFP construct successfully stained HeLa cells (Nizak et al., 2003; PMID: 14617357). The scFv-Fc fusion version of this antibody — hTA10 — detected Giantin when cells were fixed by paraformaldehyde (Moutel et al., 2009; PMID: 19245715). Precise characterization of the TA10 target was achieved; a region of 79 amino acids, representing less than 2.5% of the entire protein, was determined as its binding site (Vielemeyer et al., 2010; PMID: 20727208).
