Background: GITR (Glucocorticoid-Induced TNFR-Related protein), also known as TNFRSF18 (tumor necrosis factor receptor superfamily member18) or CD357 (Cluster of Differentiation 357), is a co-stimulatory receptor belonging to the TNF (tumor necrosis factor) receptor superfamily. It is constitutively expressed on regulatory T cells (Tregs) and is upregulated on activated CD4+ and CD8+ T cells, NK (natural killer) cells, and myeloid cells. GITR interacts with its ligand, GITRL, to modulate immune responses by enhancing effector T cell activation and proliferation while reducing Treg-mediated suppression. Due to its dual role in promoting immune activation and attenuating immune regulation, GITR is a promising target for cancer immunotherapy and autoimmune disease research.
