CloneID: HCV2
Antigen Long Description: The original antibody was generated by panning against HCV IRES construct.
Buffer Composition: PBS only.
Chimeric Use Statement: This chimeric rabbit antibody was made using the variable domain sequences of the original Human Fab format for improved compatibility with existing reagents assays and techniques.
Specificity Statement: The antibody recognizes a region located around the four-way junction structure – JIIIabc. In particular, the antibody interacts with the four unpaired nucleotides (A162, G163, U164 and A165) from the IIIa loop and an unpaired nucleotide (G233) and three base-paired nucleotides (G231, C232 and C237) from IIIc.
Application Notes (Clone): Specificity of this antibody was confirmed by ELISA tests. The antibody binds the RNA with dissociation constants of 32 ± 7 nM specifically recognizing the so-called junction IIIabc (JIIIabc). In vitro translation assay demonstrated that the antibody binds to junction IIIabc of HCV IRES inhibiting IRES-mediated translation in a concentration-dependent manner, which indicates this antibody might be used in the development of the therapeutic agents (PMID: 31765566)
