Specificity Statement: This antibody recognizes an extracellular epitope of human HER2.
Application Notes (Clone): This antibody has been successfully used to immunoprecipitate p185 erbB-2 protein from human SKBR3 breast tumor cell lysates and human erbB-2-transfected mouse HCll R111 cells. Furthermore, the binding of this antibody to surface-expressed HER2 on SKBR3 cells was determined using flow cytometry. Treatment with this antibody led to a rapid increase in the phosphotyrosine content of the erbB-2 protein in various cell lines. The binding of this antibody stimulated the turnover of the erbB-2 protein but did not stimulate DNA synthesis in HCll R111 cells. Therefore, it can be inferred that this antibody acts as a partial ligand agonist (Harwerth et al., 1992; PMID: 1353079). The binding affinity of this antibody for human HER2 was determined using ELISA (Wels et al., 1992; PMID: 1369487). The scFv version of this antibody was used in the generation of scFv(FRP5)-ETA, an antibody-exotoxin recombinant protein and tested for its anti-tumor activity (Harwerth et al., 1993; PMID: 7903153). ScFv(FRP5)-ETA displayed potent antitumoral activity in vitro against a wide range of established and primary human tumor cells, including breast and ovarian carcinomas, squamous cell carcinomas and prostate carcinomas. scFv(FRP5)-ETA also effectively inhibited the in vivo growth of human tumor xenografts and murine and rat tumor cells stably transfected with human c-erbB2 constructs (von Minckwitz et al., 2005; PMID: 16168106).
