Assay Conditions: A white 96-well plate was precoated with IL-11 protein (MedChem Express #HY-P7031) (100 ng/well). Blocking Buffer 3 (#79743) and PBST were used for blocking and washing steps, respectively. For protein titration a serial dilution of Anti-IL-11 antibody (#102663) at the highest concentration of 4 ng/µl (200 ng/reaction) was prepared in 1x PP-02 Assay Buffer (#82620). Diluted Anti-IL-11 was then added to the wells and plate was incubated at Room Temperature (RT) for 1 hour. The plate was then washed again and incubated with anti-Fc-HRP conjugate diluted 1:1000 in Blocking Buffer 3 for 1 hour at RT. After the final wash, ELISA ECL mix (#79670) was added to each well and chemiluminescence was read.
Background: Interleukin 11 (IL-11) is secreted by bone marrow stromal cells (fibroblasts) and by a number of mesenchymal cells. The IL-11 receptor (alpha chain) utilizes gp130 as its signal transducer, which is also a component of other cytokine receptors. The alpha chain of the IL-11 receptor has been identified independently as Etl2. IL-11 promotes primary and secondary immune responses in vitro and in vivo and modulates antigen-specific antibody reactions. IL-11 promotes the proliferation of IL-6-dependent plasmacytoma cell lines in the presence of neutralizing anti-IL-6 antibodies. IL-11 also stimulates the T cell-dependent development of IgG-secreting B cells in spleen cell cultures. IL-11 inhibits the differentiation of pre-adipocytes. It induces the synthesis of some acute phase proteins by hepatocytes. Monoclonal antibodies targeting IL-11 are in clinical trials, such as BI 765423 and 9MW3811, which aim to combat idiopathic pulmonary fibrosis (IPF) and cancer.
