Specificity Statement: This antibody binds human Interleukin-1 alpha, which is a cytokine constitutively present intracellularly in nearly all resting non-hematopoietic cells that plays an important role in inflammation and bridges the innate and adaptive immune systems. IL-1α is produced mainly by activated macrophages, as well as neutrophils, epithelial cells, and endothelial cells.
Application Notes (Clone): The original mouse IgG antibody binds human IL-1A with a binding affinity of Kd= 0.6 nM. The binding characterization of this antibody was done using ELISA. In vitro antibody potency was determined by its ability to inhibit IL-8 production by MRC-5 cells. This antibody was able to inhibit IL-8 production by MRC-5 cells with an IC50 of 8.90E−11M. This antibody was used in combination with 1B12.4H4 (anti-IL-1β) antibody for the generation of anti-human IL-1α/β DVD-Ig molecule that had therapeutic potential (PMID: 20068402).
