Antigen Long Description: The original antibody was generated by immunizing mice with a synthetic peptide corresponding to amino acids 5-16 (KLVVVGAVGVGK) of a mutated RAS gene encoding Val instead of Gly at position 12 coupled to the carrier protein bovine thyroglobulin (BTG) and to keyhole limpet hemocyanin (KLH).

Specificity Statement: This antibody recognizes human mutant H-, K-, and N-Ras proteins. This antibody may also react with other Ras mutants with a hydrophobic amino acid (V, A, C, H, I, L, M, Y, T, N, Q) at position 12 (PMID: 9440050). This ras protein binds GDP/GTP and possess intrinsic GTPase activity, that results in conversion of nucleotide guanosine triphosphate (GTP) into guanosine diphosphate (GDP). This protein plays an important role in many signal transduction pathways and regulation of cell proliferation. Somatic ras mutations are highly observed in leukemias, colorectal cancer, pancreatic cancer and lung cancer.
Application Notes (Clone): This antibody was capable of binding G12V and G12C mutant peptides of human ras protein in competition ELISAs (PMID: 3094010). It was reported to cross react with peptides containing amino acids V, A, C, H, I, L, M, Y, T, N, Q at position 12 in an ELISA (PMID: 9440050). This antibody could detect mutant KRAS proteins from cell lysates of human tumor cell lines in a western blot, but did not react with normal KRAS or other KRAS mutants. Immunohistochemical studies showed that DWP-labeled transformed NIH cells and human carcinoma cells contained p21ras is with either Val or Cys at position 12 but not normal or other activated KRAS protein (PMID: 3094010).
