Specificity Statement: The antibody is specific for human-RANK-L. RANKL is a tumor necrosis factor related protein that binds to, and activates, the TNF receptor related protein Receptor Activator of NF-kB (RANK). RANKL acts as the key cytokine that regulates osteoclast differentiation and activation during normal bone remodeling and during disease
Application Notes (Clone): The affinity of the original format of the antibody is 0.17x 10-12 nM, as measured by PAE technology. RAW264.7, a murine macrophage cell line that can be induced by RANKL to differentiate into osteoclast-like cells, was used in a TRAP test. In the TRAP test, the original antibody or the Fab fragment were able to neutralize the stimulating effect of RANKL on osteoclast formation from a RAW 264.7 culture. Further, the addition of the original format of the antibody or the Fab fragment inhibited osteoclast differentiation of the RAW cell in a dose-dependent manner. The affinity of the original format and the Fab fragment of the antibody was measured by the Scatchard method (affinity = 8.7 and 6.5 pM). The original antibody and the Fab fragment could significantly increase the bone densities and bone calcium content in a dose-dependent manner, in in vivo experiments (WO2011116527A1).
