Specificity Statement: The antibody specifically binds human RSPO1. The antibody cross-reacts with mouse RSPO1
Application Notes (Clone): The binding affinity of the original format of the antibody to human and mouse RSPO1 was measured by surface plasmon resonance (Kd less than 0.1 nM respectively). Further, the binding affinity of the humanized version of the antibody, h89M5-H2L2, to human RSPO1 was measured by surface plasmon resonance (Kd less than 0.1 nM). The original format of the antibody could inhibit tumor cell growth in vivo in an ovarian tumor xenograft model as single agents and in combination with a chemotherapeutic agent. The original format of the antibody could inhibit tumor cell growth in vivo in a pancreatic tumor xenograft model in combination with a chemotherapeutic agent. The original antibody was able to block RSPO1-induced β-catenin signaling in HEK-293 cells. The original antibody blocked binding of RSPO1 to LGR5 in HEK-293 cells, as measured by FACS. The original and humanized format of the antibody, could bind to human RSPO1 expressed on HEK-293 cells, as shown by FACS analysis (US8802097B2).
