CloneID: 8D4
Heavy Chain modification: Fc Silent™
Antigen Long Description: The original antibody was generated by immunizing BALB/c mice with purified cytoskeletal Talin, from chicken gizzard.
Buffer Composition: PBS with 0.02% Proclin 300.
Chimeric Use Statement: This is a reformatted human IgG1 Fc Silent™ antibody, based on the original human IgG1 format, created for improved compatibility with existing reagents, assays and techniques.
Available Custom Conjugation Options: AP, HRP, Fluorescein, APC, PE, Biotin Type A, Biotin Type B, Streptavidin, FluoroProbes 647H, Atto488, APC/Cy7, PE/Cy7
Uniprot Accession No.: Q9Y490
Specificity Statement: The antibody is specific for mammalian and avian talin. Talin is a multifunctional constituent of cell-substratum attachment sites and it has a high molecular weight (225-235 kDa). The protein is found in a variety of tissues and cell types.
Application Notes (Clone): The antibody detected the N-terminal region of talin1 by western blot analysis (Praekelt et al., 2012; PMID: 22306379 and Martel et al., 2001; PMID: 11279249). However, it also detected the equivalent region of talin2, albeit to a lesser degree (Praekelt et al., 2012; PMID: 22306379). This antibody was used for detection of talin expression on HeLa/AT22, AT21, wild-type HeLa cells or pMEP4-transfected HeLa cells cells by flow cytometry (Albigès-Rizo et al., 1995; PMID: 7593292). lmmunocytochemistry using the antibody showed patterns of talin expression within DRG growth cones (Sydor et al., 1996; PMID: 8794861). Indirect immunofluorescence staining of cultured fibroblasts (rat, human, chicken and xenopus embryo fibroblasts) was tested with the antibody. In all the samples the focal adhesions were stained with the antibody (Otey et al. 1990; PMID: 1690175). Talin was immunoprecipitated from MDA-MB-231 cell lysates using the antibody (Beaty et al., 2014; PMID: 24891603).
