Specificity Statement: This antibody binds human tissue factor (TF) a hemostatic protein which is involved in the extrinsic pathway of coagulation. TF is receptor for circulating serine protease factor VIIa, activating the catalytic VIIa protease domain. The complex formed functions to activate the zymogen factor X into activated factor Xa, which then catalyzes the formation of Prothrombin to Thrombin. The pathways results in a fibrin clot which acts to stop bleeding. TF plays a role in normal hemostasis by initiating the cell-surface assembly and propagation of the coagulation protease cascade.
Application Notes (Clone): The binding characterization of this Fab antibody was done using ELISA and dot blot. This antibody shows positive plasma coagulation inhibitory activity. This antibody is capable of neutralizing human tissue factor in vitro by inhibiting the binding of TF factor/Vlla complex to factor X. The inhibition rate increase with the concentration. The pharmacodynamic and pharmacokinetic activity of this antibody was studied in vivo in rat models (CN101423552).
