Assay Conditions: A white 96-well plate was precoated with TROP-2, His-Tag protein (AcroBiosystems #TR2-H5223) (100 ng/well). Blocking Buffer 3 (#79743) and PBST were used for blocking and washing steps, respectively. For protein titration a serial dilution of Anti-TROP2 Antibody, Biotin-Labeled (#102729) at the highest concentration of 4 ng/µl (200 ng/reaction) was prepared in 1x PP-02 Assay Buffer (#82620). Diluted Anti-TROP2 was then added to the wells and plate was incubated at Room Temperature (RT) for 1 hour. The plate was then washed again and incubated with anti-Fc-HRP conjugate diluted 1:1000 in Blocking Buffer 3 for 1 hour at RT. After the final wash, ELISA ECL mix (#79670) was added to each well and chemiluminescence was read.
Background: TROP2 (Trophoblast cell-surface antigen 2), also referred to as tumor associated calcium signal transducer 2 (TACSTD2), GA733-1 or M1S1, is a cell surface glycoprotein that is highly expressed in a variety of solid cancers yet has low expression in normal cells. Through a variety of signaling pathways, such as the MAPK (mitogen-activated protein kinase)/PI3K (phosphoinositide 3-kinase)/AKT (protein kinase B) pathway TROP2 regulates cancer growth and metastasis. Overexpression of this protein is associated with a poor prognosis in cancers such as breast, gastric and ovarian cancer. In other cases, such as NSCLC (non-small cell lung cancer) lower expression seems to link to malignancy. TROP2 is a favorable target for antibody drug conjugates (ADC) and immunotherapy. Studies with Sacituzumab govitecan, an ADC composed of an anti-TROP2 humanized monoclonal antibody and SN-38 (topoisomerase I inhibitor), in cell models have shown promising results.

This anti-TROP2 antibody is a purified, humanized recombinant monoclonal antibody, which contains an Avi-Tag™ and is biotinylated at the C-terminus of the heavy chain unit. This antibody has been tested in a binding assay. This antibody is similar to Sacituzumab.
