Application: Useful for studying the expression of human TCR γδ on the cell surface by flow cytometry.
Assay Conditions: One million frozen cells from a) Normal PBMCs (#79059), b) Untransduced T Cells (#78170), c) Expanded Human Peripheral Blood γδ T Cells (Vδ1) (#82443), and d) Expanded Human Peripheral Blood γδ T Cells (Vγ9Vδ2) (#82733), were thawed, blocked, and stained with 1 µg of Fluosite™ Anti-PAN TCR γδ Antibody, PE-Labeled (#102777) and APC anti-human CD3 Antibody (Biolegend #344811) for 30 minutes on ice. Samples were then washed three times and analyzed by flow cytometry. The X axis represents PE intensity, while the Y axis represents APC intensity. Quadrant 2-2 (Q2-2) of the density plots represent the percentage of CD3+ and TCR γδ+ cells. Each plot was gated on FSC-A/SSCA (to remove debris from analysis) and FSC-H/FSC-A (singlet discrimination) (not shown).
Background: Unlike conventional αβ T cells, γδ T cells contribute to innate and adaptive immune responses and are primarily involved in tumor surveillance, wound healing, and mucosal immunity. An anti-PAN TCR γδ antibody specifically binds to a conserved epitope shared among most human γδ TCR variants, making it a valuable tool for identifying and characterizing γδ T cells across various tissues and disease contexts. This reagent is widely used in flow cytometry and immunophenotyping studies to investigate the functional roles of γδ T cells in health and disease.
Cross Reactivity: This antibody recognizes human TCR γδ. It has not been tested in other species.
Description: This anti-PAN TCR antibody is a purified monospecific recombinant antibody, which is labeled with R-Phycoerythrin (PE). This antibody has been tested by flow cytometry for binding to two major subsets of γδ TCR (T cell receptor), Vδ1 TCR and Vδ2 TCR, in normal human PBMCs (Peripheral Blood Mononuclear Cells, BPS Bioscience #79059), Expanded Human Peripheral Blood Gamma Delta T Cells (Vδ1; BPS Bioscience #82443), Expanded Human Peripheral Blood Gamma Delta T Cells (Vγ9Vδ2; BPS Bioscience #82733). Untransduced T Cells (BPS Bioscience #78170) were used as a negative control, as they express only αβ TCR. FluoSite™ is a tag-based method for site-specific conjugation of antibodies with fluorophores, ensuring the labeling site is far from the antigen-binding domain to preserve antibody functionality. This precise, consistent labeling ensures the homogeneity of the labeled antibody preparation, making it ideal for flow cytometry and other high-sensitivity applications.
