Application: Useful for binding assays.
Assay Conditions: A 96-well plate was coated with RNF43, Avi-His-Tag Recombinant (100 ng/well in PBS) at 4°C overnight. The plate was washed three times with PBST, and 200 µl Blocking Buffer 3 was added for 2 hours at room temperature (RT). After three washes in PBST, a 1:2 serial dilution of RSPO2-Fc protein prepared in PP-02 Assay Buffer (#82620), was added and incubated at RT for 1 hour. The plate was washed three times and incubated with Anti-human Fc-HRP conjugate diluted of 1:1000 in Blocking Buffer 3 for 1 hour at RT. After three washes, 100 µl of the ELISA ECL Substrate (#79670) was added to each well and chemiluminescence was measured.
Background: RNF43, or ring finger protein 43, is an E3 ubiquitin ligase that acts a tumor suppressor to negatively regulate the WNT (wingless-related integration site) signaling pathway through promoting the degradation of frizzled WNT receptors. Loss of function mutations in RNF43 have been shown to be a predictive biomarker in favorable response to anti-BRAF (B-Raf proto-oncogene serine/threonine kinase)/EGFR (epidermal growth factor receptor) therapies in colorectal cancer patients. RNF43 mutations are frequent predictors, and therefore of high clinical interest, in pancreatic, gastric, and endometrial cancers. Further studies will reveal more on the cancer signaling pathways and combinatory therapies involving RNF43.
