Heavy Chain modification: Fc Silent™

Antigen Long Description: The original antibody was generated by immunizing mice with recombinant human CD40 protein.

Buffer Composition: PBS with 0.02% Proclin 300.

Chimeric Use Statement: This chimeric human antibody was made using the variable domain sequences of the original Mouse IgG1 format for improved compatibility with existing reagents assays and techniques.

Available Custom Conjugation Options: AP, HRP, Fluorescein, APC, PE, Biotin Type A, Biotin Type B, Streptavidin, FluoroProbes 647H, Atto488, APC/Cy7, PE/Cy7

Uniprot Accession No.: P25942

Specificity Statement: This antibody recognizes and binds human and primate CD40, but does not cross react with mouse CD40. CD40 acts as a receptor for TNFSF5/CD40LG. This protein is a receptor on antigen-presenting cells of the immune system and is essential for mediating a broad variety of immune and inflammatory responses including T cell-dependent immunoglobulin class switching, memory B cell development, and germinal center formation.

Application Notes (Clone): The reactivity of this antibody to human, Ganlamp monkey and mouse CD40 protein expressed on the surface of CHO cells was determined using flow cytometry. This antibody positively binds human and primate CD40 proteins and does not cross react with mouse CD40. The original mouse IgG antibody binds human CD40 with a binding affinity of kd= 6.13X10^(-10) M. The tumor inhibitory activity of this antibody was also tested in vivo in B-HCD40 mice injected with murine MC-38 cells (colon cancer cells). This antibody exhibited good tumor inhibitory effect (CN111763259).
