Specificity Statement: This antibody binds an epitope called ANK-1 present on most NCAM isoforms expressed on a subset of NK cells of mouse and human. This epitope is selectively expressed on pre-activated, adherent natural killer NK cells (pre-A-NK cells), which are interleukin (IL)-2-stimulated NK cells, that are selectively endowed with the increased expression of integrins and ability to adhere to solid surfaces, migrate into, infiltrate, and destroy cancerous tissues.
Application Notes (Clone): This antibody was used to classify the nonactivated human peripheral blood NK cells expressing ANK-1 epitope using flow cytometry. It was reported that ANK-1 epitope was preferentially expressed by around 26% of NK cells (range 9–56%) and was exclusively CD56dim. This antibody was also capable of detecting ANK-1 epitope from crude human peripheral blood NK cell and affinity purified brain N-CAM lysates using western blot. This antibody was also used to test the ability of NK cells to respond to IL-2 by generation and expansion of A-NK cells with LAK activity, following depletion of ANK-1+ NK cells by treatment with mAb ANK-1 and complement. This antibody can also be used for the immunohistochemical staining of paraformaldehyde fixed paraffin embedded tissues of spleen, lymph nodes, tonsils, intestines, lungs, and liver tissues (PMID: 15356097).
