Specificity Statement: The antibody is specific for HB-EGF.
Application Notes (Clone): The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody detected the recombinant secretory human HB-EGF by western blot analysis. The HB-EGF-EGFR binding inhibitory activity of the original antibody was examined using 32D/EGFR cell and biotin-labeled HB-EGF. The original antibody concentration-dependently inhibited binding of biotinylated HB-EGF to EGFR. Further, it could also inhibit binding of HB-EGF with EGFR. The neutralizing activity of the original antibody was examined by a cell growth inhibition assay which used an HB-EGF-dependent cell. The original antibody showed HB-EGF-dependent cell growth inhibition activity for the cell line 32D/EGFR. The original antibody bound to the membrane type and cell membrane-bound HB-EGF of cancer cell lines (human gastric cancer cell line MKN-28, human ovarian cancer cell line ES-2, human breast cancer cell line MDA-MB-231). The binding affinity of the original format of the antibody to human HB-EGF was measured by surface plasmon resonance (Ka= 2.7 1/Ms). The original antibody specifically bound to the T cell leukemia and acute myelogenous leukemia cell lines expressing HB-EGF as shown by FC (US7851601B2).
