Specificity Statement: This antibody binds human Interleukin-1 beta, which is a potent pro-inflammatory cytokine. Initially discovered as the major endogenous pyrogen, induces prostaglandin synthesis, neutrophil influx and activation, T-cell activation and cytokine production, B-cell activation and antibody production, and fibroblast proliferation and collagen production. It promotes Th17 differentiation of T-cells and synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells. It also plays a role in angiogenesis by inducing VEGF production synergistically with TNF and IL6.
Application Notes (Clone): The original mouse IgG antibody binds human IL-1B with a binding affinity of Kd= 0.26 nM. The binding characterization of this antibody was done using ELISA. In vitro antibody potency was determined by its ability to inhibit IL-8 production by MRC-5 cells. This antibody was able to inhibit IL-8 production by MRC-5 cells with an IC50 of 5.30E−10M. This antibody was used in combination with 18F4.2C8 (anti-IL-1α) antibody for the generation of anti-human IL-1α/β DVD-Ig molecule that had therapeutic potential (PMID: 20068402).
