Specificity Statement: The antibody binds to human and rhesus macaque LAG-3.

Application Notes (Clone): The specificity of the original format of the antibody was confirmed by ELISA analysis. The antibody was able to bind LAG-3 expressed at the surface of HEK-293 cells was evaluated by flow cytometry. The antibody showed binding to activated T cells. The antibody bound to cells expressing rhesus LAG-3 on transiently transfected HEK-293 cells. The affinity of the antibody to human LAG-3 was measured using Bio-Layer Interferometry (Kd = 0.70 nM). The antibody showed inhibitory capacity of the binding of LAG-3 to MHC class II in a dose dependent manner, and was able to completely block binding of LAG-3 to MHC-II at high concentrations (IC50 = 12 nM). The antibody could block the binding of LAG-3 to MHC class II-expressing Daudi cells in a dose-dependent manner by flow cytometry analysis. The antibody was able reverse exhaustion and restore the activity of T cells to secrete IL-2 and IFN-γ upon restimulation at high doses (US20190040136A1).
