Specificity Statement: This antibody binds human PAI-1 at an epitope between amino acids 277 and 327, more particularly it binds residues between Arg300, Gln303 and Asp305 located on the loop between α-helix I and β-strand 5A. This antibody cross-reacts with pig PAI-1 but not with mouse or rat. Plasminogen activator inhibitor-1 (PAI-1), is a serine protease inhibitor, a unique member of the serpin superfamily which plays an important role in fibrinolysis and is an established risk factor for cardiovascular diseases. It a primary inhibitor of tissue-type plasminogen activator (PLAT) and urokinase-type plasminogen activator (PLAU). As PLAT inhibitor, it is required for fibrinolysis down-regulation and is responsible for the controlled degradation of blood clots. As PLAU inhibitor, it is involved in the regulation of cell adhesion and spreading .
Application Notes (Clone): This antibody is reported to have strong inhibitory effect towards PAI-1 activity. The cross reactivity of this antibody towards other PAI-1 than human was done using ELISA (PMID: 9048903). This antibody shifts the reaction to the substrate pathway without decreasing the rate-limiting constant for reactive center loop insertion by tPA (PMID: 12223472). This antibody interferes with the final locking step in the serpin/proteinase interaction, thereby explaining its substrate inducing properties (PMID: 12871373).
