CloneID: AbAb1-PLCy
Antigen Long Description: The original antibody was generated by immunizing rabbits with recombinant phosphorylated peptide pPLC-γ(RNPGF-pY-VEANPGGGSC) conjugated with KLH as an immunogen.
Buffer Composition: PBS with 0.02% Proclin 300.
Available Custom Conjugation Options: AP, HRP, Fluorescein, APC, PE, Biotin Type A, Biotin Type B, Streptavidin, FluoroProbes 647H, Atto488, APC/Cy7, PE/Cy7
Uniprot Accession No.: P19174
Specificity Statement: This antibody binds phosphorylated phosphoinositide phospholipase C-gamma, which mediates the production of the second messenger molecules diacylglycerol (DAG) and inositol 1,4,5-trisphosphate (IP3). It also plays an important role in the regulation of intracellular signaling cascades.
Application Notes (Clone): The binding reactivity of this antibody to recombinant phosphorylated pPLC-γ peptide was confirmed using ELISA. The binding reactivity was further confirmed using western blot, of phosphorylated PLC-γ from NIH3T3 cell lysates. The scFv version of this antibody was used fused to human IgG1 using a suitable linker for the generation of scFv-Fc mini-antibody. This minibody was capable of immunoprecipitating pPLC-γ. The fluorescent scFv version of this antibody, when expressed intracellularly was reported to inhibit signal transduction by PLC-γ in transfected into NIH3T3 cells as observed using fluorescent microscopy (US20090233358).
