Corning with its brands Corning, Costar, Falcon, Axygen and MediaTech is the specialist in the field of cell culture. In addition to the usual cell culture-treated surfaces that Corning offers in all common formats such as culture plates, dishes and flasks, some more special cell culture surfaces are available. The best examples are the CellBIND and Primaria surfaces for cells that are difficult to adhere as an alternative to coatings. Untreated products and an ultra-low attachment surface are also available. Matrigel products come from Falcon. Corning also offers roller and spinner bottles, flasks and transwell systems.

A very popular product from Falcon are the Cell Strainers, which fit perfectly into the Falcon 50mL tubes. They are color-coded and available with a nylon membrane with mesh sizes 40µm, 70µm and 100µm. The Cell Strainers are sterile and individually packed.

The BioCoat cell culture products from BD represent a unique product line. They combine the Falcon multiwell plates, dishes, flasks, culture slides and glas coverslips with the extracellular matrices. Using a special manufacturing process, the ECM components are attached to the surfaces and bound. This results in a uniform, optically clear matrix. Due to quality controls, a guaranteed performance of each lot and lot-to-lot consistency is given. The BioCoat products promote cell attachment, spread and growth of cells as well as the differentiation of primary cells and cell lines in serum-containing and serum-free cultures.

A special product is the HYPERFlask. The name is derived from high yield performance.

This flask is ideal for the production of a large number of cells because it consists of 10 identical layers within the flask and thus provides 10 times the growth surface of a normal T175 bottle. And that with the same external dimensions. The HYPERFlask can be used both automated and manually. All layers are filled at the same time through one opening. Each layer consists of a gas-permeable polystyrene surface that has been treated like the CellBIND cell culture surface. There are gaps between the individual layers for gas exchange.

The Falcon Culture Slides are a safe and easy way to provide separate compartments on a slide for microscopic analysis. They are available in a 4-well and a 8-well version.

Falcon's extracellular matrices, known as Matrigel, are another important line of products. The interaction of cells with the basal membrane is an important factor in regulating cell behaviour. In vivo conditions can be simulated very well by using Matrigel in vitro.

BD Biosciences use a patented technology to extract the Matrigel matrix from EHS mouse sarcoma, a tumor rich in extracellular matrix proteins. The Matrigel matrix consists of laminin, collagen IV, entactin and heparin sulfate proteoglycan. Further components are growth factors, matrix metalloproteinases and others. At 4°C it is a solution, at room temperature a three-dimensional structure is reconstituted. This mimics the in vivo conditions at the basement membrane in terms of physical and functional view.

Matrigel is particularly suitable for epithelial cells. It promotes the differentiation of hepatocytes, SMC, glandular cells, neurons, etc.

The cells behave more or less as if in vivo conditions. The Matrigel Matrix thus creates the possibilities for studies with regard to the morphology of cells, biochemical functions, migration or invasion and gene expression.

The new RAFT system from Lonza simulates the natural environment of cells as closely as possible. The kits contain everything you need for your 3D cell culture. The system uses rat tail collagen type 1 and the patented absorber technology to create a realistic cell environment in less than an hour.

The versatile RAFT kit is available in different formats and enables examinations using imaging, biochemical and histological techniques. In connection with the primary cells and the special media from Lonza, the RAFT system enables the development of physiologically relevant cell culture models. The RAFT kits have already been successfully tested with many cell types in various research areas, including oncology, toxicology and "barrier modeling".

 

WHAT IS THE RAFT SYSTEM COMPOSED OF?

The reagent kit contains rat tail collagen type 1 as an extracellular matrix and a special neutralizing solution. With this solution, the neutralization can be carried out in one step. This is an advantage compared to lengthy, error-prone titration methods. A 10x MEM is also included as a medium.

Another important component is the RAFT absorber. The biocompatible absorbers work in 24-well and 96-well format. After the cells are mixed with the matrix, excess liquid is removed with the absorber and the matrix is ​​compacted. The result is the production of "high-density" collagen matrices under controlled conditions, in which the desired cell types are embedded. Such “frameworks” represent a natural, in vivo-like environment for cells and thus enable their growth and interaction.

Another variant is the RAFT kit with the 24-well inserts for use with permeable inserts. Thus, RAFT can also be used in co-culture models and "barrier models and air-lift models".

RAFT 3D cell culture systems have been successfully tested in BioTek applications with the multi-mode imager Cytation 1 and Cytation 5 .
