Applications: Useful for studying expression of CD3 on the cell surface by flow cytometry. For flow cytometric staining, the suggested use of this reagent is 5 µl (1 µg antibody) per million cells in 100 µl staining volume.
Assay Conditions: One million PBMCs (BPS Bioscience #79059) were blocked and stained with 1 µg of anti-CD3 antibody, FITC-labeled (BPS Bioscience #102008) or FITC Human IgG1 Isotype Control (BioLegend #403507) for 30 minutes on ice, washed three times, and analyzed by flow cytometry. Non-lymphoid cells were excluded from the analysis based on side scatter and forward scatter signals. Y-axis represents the cell count, while X-axis shows FITC intensity.
Background: CD3 (cluster of differentiation 3) is a complex made of CD3γ chain, a CD3δ chain and two CD3ε chains. CD3 associates with the TCR (T-cell receptor) and a CD3ζ chain to activate cytotoxic T Cells (CD8+) and T helper cells (CD4+) and form the TCR complex. It is found in T cells, during all developmental stages, and it is a typical T cell marker. Its essential role in T cell activation has made CD3 an attractive and useful molecule for the development of anti-cancer drugs. For example, CAR-T cells (chimeric receptor antigen T cells) made use of the CD3ζ domain for intracellular signaling activation.
Cross Reactivity: This antibody recognizes human CD3. It has not been tested on other species.
