Primary cells are cells directly isolated from human or animal healthy tissues and that reflect the biological diversity of the different body districts and donors they are isolated from.
Why using primary cells?
To produce biologically relevant data for publication or drug discovery/development you need to choose the best model (combination of the right cells/ right tissue/ right donor). Compared to immortalized cell lines, primary cells are a better and more biologically relevant model as they better reflect in-vivo conditions. Furthermore primary cells represent the biological variation within a population and have limited amount of accumulated mutations.
Why using cell lines?
Cell lines are isolated from tumors or immortalized to be maintained in culture under artificial conditions, and may be used with limitations to study basic cell behavior. Since tumor cells differ significantly from healthy cells, the use of cell lines to study normal cell behavior is harshly criticized.
